Mechanism of action
- FOXO4-DRI is a synthetic D-retro-inverso peptide (mirror-image design for higher stability) that disrupts the interaction between the transcription factor FOXO4 and the tumour suppressor p53.
- In senescent ("aged") cells this interaction retains p53 in the nucleus and prevents their self-destruction.
- By disrupting it, p53 is intended to be released and selectively trigger apoptosis of senescent cells (a senolytic approach).
- Healthy cells are intended to be largely spared.
Research status
- FOXO4-DRI has so far been studied exclusively preclinically.
- A widely noted study (2017) described the selective elimination of senescent cells in animal models; more recent studies investigate applications such as keloid fibroblasts or age-related testosterone insufficiency in animal models.
- Controlled human studies do not exist.
- Safety and efficacy in humans are not demonstrated.

Frequently asked questions
What is FOXO4-DRI?
FOXO4-DRI is a synthetic D-retro-inverso peptide (mirror-image design for higher stability) that disrupts the interaction between the transcription factor FOXO4 and the tumour suppressor p53.
What type of peptide is FOXO4-DRI?
FOXO4-DRI belongs to the Other class.
Is FOXO4-DRI legal in Germany?
FOXO4-DRI is not approved as a medicine in Germany. Obtaining it outside clinical studies is legally problematic.
Where can I buy FOXO4-DRI?
Peptica sells nothing and names no sources. FOXO4-DRI is "not approved" in Germany; obtaining unapproved substances outside clinical studies is illegal and carries quality and safety risks.
Is FOXO4-DRI banned in sport (WADA)?
FOXO4-DRI is currently not on the WADA Prohibited List.
What are the known risks of FOXO4-DRI?
Documented risks include: No human studies — safety profile entirely unknown; Interference with p53/apoptosis signalling with potentially far-reaching consequences. This is not a complete safety assessment and not medical advice — see the risks section for the full list.
How well researched is FOXO4-DRI?

Legal assessment
FOXO4-DRI is not approved as a medicine and is solely an experimental research substance. Not on the WADA Prohibited List. Interfering with p53 signalling is potentially risky; obtaining it as a research chemical is highly risky.
